Antibiotics susceptibility patterns, plasmid profiles, and restriction enzyme analysis of plasmid DNA were used in the epidemiological typing of Shigella sonnei isolated in Korea, 1993 and 1994. In the antibiotic susceptibility tests, 34 of 48
Shigella
sonnei isolates showed resistance to streptomycin, tetracycline, rifampin, and sulfamethoxazole-trimethoprim. Most of the examined isolates exhibited a multiresistancc to antibiotics. A total of 46 of 48 S. sonnei was piaced into 5 plasmid
patterns.
72.9% of S. sonnei isolates were included in SI pattern which showed 1.7, 2.0, 3.4, 5.1, and 8.6kb plasmid. The common size of plasmid among 5 groups was 2.0 kb. S. sonnei KNIH104S included into SI group was used to construct vaccine candidate.
The
vaccine candidates (LMG1 to LMG6) were made by P1 phage transduction of aroL gene from E. coli to S. sonnei KNIH104S. They showed sereny-negative and 50% protection rate in guinea-pigs against challenge with the virulent wild-type strain
KNIH104S.
